چکیده

The pathway of de novo pyrimidine biosynthesis in the amastigote and Promastigote forms of Leishmania tropica has been investigated. Both parasite forms contained all six enzyme activities. Carbamoyl phosphate synthase (CPSase) utilizes Lglutamine and not ammonia as the amine group donor. Dihydroorotate dehydrogenase (DHO-DHase) was found to be cytoplasmic, where as orotate phosphoribosyltransferase (OPRTase) and orotidylate decarboxylase (ODCase) were found to be particulate. N(phosphonacetyl)-L-aspartate, dihydro-5azaorotate, 5-azaorotate and 6-azaUMP were found to be a potent inhibitor of the aspartate transcarbamoylase (ATCase), DHO-DHase, OPRTase, and ODCase, respectively.

Aspartate Transcarbamylase from Leishmania donovani

Leishmania donovani is a protozoal pathogen that belongs to the kinetoplastida order. Unlike in other eucaryotic systems, the first three enzymes of the de novo pyrimidine biosynthetic pathway are not components of a multifunctional protein system. The three enzyme activities in the crude extract were separated on a Sephacryl S-200 column. Aspartate carbamoyltransferase (EC 2.1.3.2) has been pu...

Insights into the pyrimidine biosynthetic pathway of human malaria parasite Plasmodium falciparum as chemotherapeutic target.

Malaria is a major cause of morbidity and mortality in humans. Artemisinins remain as the first-line treatment for Plasmodium falciparum (P. falciparum) malaria although drug resistance has already emerged and spread in Southeast Asia. Thus, to fight this disease, there is an urgent need to develop new antimalarial drugs for malaria chemotherapy. Unlike human host cells, P. falciparum cannot sa...

Malaria Parasite Pyrimidine Nucleotide Metabolism: A Promising Drug Target

Malaria is a major cause of morbidity and mortality in the tropical and subtropical endemic countries worldwide. This is largely due to the emergence and spread of resistance to most antimalarial drugs currently available, including the first-line treatment artemisinins. Thus, to fight this disease, there is an essential requirement to develop new antimalarial drugs for malaria chemotherapy. Pl...

The Pyrimidine Nucleotide Biosynthetic Pathway Modulates Production of Biofilm Determinants in Escherichia coli

Bacteria are often found in multicellular communities known as biofilms, which constitute a resistance form against environmental stresses. Extracellular adhesion and cell aggregation factors, responsible for bacterial biofilm formation and maintenance, are tightly regulated in response to physiological and environmental cues. We show that, in Escherichia coli, inactivation of genes belonging t...
